I want to use synthesized gene for cloning and expression, instead of PCR cloning. Can any one help what are the precautionary measure to design gene sequence for synthesized gene cloning.
The gene synthesis companies will do everything for you. What you need to do is to tell them you gene coding sequence, what the expression host you will use, what the restriction sites you need if you want to re-clone the synthesised gene late, they will design the gene coding sequence with the optimized codons, cloned into their vector for you. You can provide your own vector but this service usually with an extra charge.
You can also ask most companies to codon-optimise your cDNA for the type of animal your cells are from. This can increase protein expression significantly.
