I have tried to co-express two reporters in yeast, one in TetOff system and another one from YCp50 vector, which has Tet promoter. In theory it should be fine as even if tTA binds to the Tet promoter it should only activate transcription. Does anybody know if in practice it is right or wrong? I myself saw huge downregulation or the reporter from YCp50, but this can be as well due to number of other reasons.
