where Ea is the extinction coefficient observed for the absorption band at a given DNA concentration, Ef is the extinction coefficient of the complexes in its free form and Eb of the complex in the fully bound form.

K is the binding constant

C is the concentration of drug

s is binding site size

and

B = 1 + KC + K [DNA] / 2s
