Is it possible that the protein will not be expressed by changing only one amino acid of the target protein?

Currently, we are expressing low molecular weight antibodies of about 15 kilodaltons in Escherichia coli, but by mutating only one amino acid, the expression is markedly lower compared to WT.Is mutation of 1 amino acid considerably influenced in protein expression in the cells?Any explanations or sugggestions?
