I need a transient assay of my gene in pBI121, but I don’t want to use Agrobacterium. On the other hand, can circular plasmids be expressed in plants? Can RNA polymerase II recognize 35S promoters in pBI121?
Hi, it might be possible but you have to deliver the plasmid to the cell and this could be by bombardment or by protoplast transformation but to prepare the protoplast is not easy task. Also RNA pol II recognize 35S in plant cells.
The circular plasmids can be expressed in plant cells but the gene of interest has to be under the control of appropriate promoter adn you need to deliver the plamsid to the cell. In fact whe you use protoplast transformation or bombardement you use circular plasmids. Why don't you use Agrobacterium mediated transient expression? I don t know which species you are using but it is working quite well for me.Good luck!
Yes, pBI121 can express gene-of-interest without using Agrobacterium. Two examples are here:
(1) pBI121 expression using particle bombardment in plant cells (see attached paper #1).
(2) pBI121 expression through PEG-mediated genetic transformation in rice plant cells (see attached paper #2).
However, I don't know if you are aware of a technology called 'Agroinfiltration'. For transient assay, actually Agroinfiltration is a fairly easy technology. The steps include cloning your gene into a binary vector, transferred the vector into Agrobacterium, inject the culture of Agrobacterium into for example tobacco leaves, and after 1 or 2 days you can transiently assay your gene product. It doesn't need really fancy devices to conduct. Bombardment needs a gene-gun machine. Protoplast transformation need to prepare the cell-wall protoplast, a tedious work. Besides, the binary vector (with T-DNA elements) are suitable for using in particle bombardment and PEG-mediated protoplast genetic transformation in the future, if needed.
