I am using the MethylFlash Global DNA Methylation (5-mC) ELISA Easy Kit (Colorimetric) (Base Catalog #P-1030) for one of the analyses for my doctoral thesis chapters.

In the kit manual is written that "it is advised to run replicate samples to ensure that the signal generated is validated". However, some of my DNA samples don't have enough DNA to do the second replicate (those are the only DNA samples I have, and I cannot collect them again).

I was wondering if: is it still okay to skip the replicate for the samples in which the DNA amount is not enough to run the replicate?

I would really appreciate it if you answer my inquiry fast since my thesis submission deadline is approaching, and I need your help.

Thank you so much in advance for your time and consideration.

Sincerely yours,

Jun Ishida
