I am currently planning to work on cell cycle arrest assays. I saw different literature have slightly different methods on cell synchronization. Some used serum free media, while some used 1%FBS, and some even did not perform this step. What would be the outcome of these different methods? and is it necessary to synchronize cells? Thanks
The significance of synchronization of the cell cycle is that a large number of cells in the same cycle can be obtained so that biochemical analysis can be performed and the structure and function of cells at different stages of each cell cycle can be deeply understood.
As well as studying the expression and regulation of related genes in the cell cycle, the effect of not synchronizing the effects is not significant.
Hello Nathaniel, Snow has explained the significance of synchronization already. However, if you are not interested in the kinetic analysis of the cell cycle progression, there will be little or no need for synchronization. Three distinct methods are available in literature for cell synchronization; serum deprivation, cell contact inhibition and centrifugal elutriation of cells in any phase of the cell cycle. If you are interested in knowing the outcomes of the different methods highlighted, I suggest you set up those experiments. There couldn't be a stronger convinction than the one you arrived at yourself. For details, please see the attached.
