Is it necessary to digest the livers from embryos in collagenase solution for hepatocytes preparation in order to carry out EROD measurement?

I found one literature that livers from embryos were pooled and digested for 45 min in 75 mL of a 0.5 mg/mL collagenase (Sigma) solution.

Actually what is the purpose of adding collagenase solution? Is there any alternative way for preparation?

Usually collagenase treatment is used for the preparation of cell cultures - I presume you do not want to culture the cells ? Therefore simply homogenize the liver pool in a suitable buffer for sub-cellular fractionation and isolate either microsomes of 'post-mitochondrial supernatant' depending on equipment availability
