As the presence of water may be essential for the stability of the compound.... So I am asking....
The lipids form vesicles in presence of water due to hydrophobic effect. In other word water is the driving force for vesicle formation. In organic solvent the vesicle will simply dissolve out. Do you expect micelle formation in absence of water?
Water is necessary in the core. In absence of water it will be a vacuum like situation, which is unstable, and will lead to collapse of vesicle. As a result during characterization cryo-SEM/TEM is preferred over normal SEM/TEM.
Exactly this answer was I want. Actually, i had done TEM(Normal) for beta-Cyclodextrin and Ionic liquid based anionic surfactant.
May I know where cryo-SEM/TEM & ITC will be reliable? Any institute or university? Thank you so much.
Another important question is that how to interpret such results from normal TEM?
I think you can still work with normal TEM. I personally did size distribution from normal FEG TEM. Anyway you should consult an expert.
