I am planning to carry out a miRNA expression study on my invitro cell model. Initially I had planned to proceed forward by extracting the total RNA, small RNA library prep and then proceed to sequencing. 

Some literature suggests that it might be advisable to do an AGO pull down assay to enrich my miRNA population before sequencing to get relevant hits and mature miRNAs in the datasets. 

I would like to know is this step really essential as the downstream data analysis part does cover the mature miRNA selection part.
