I have produced polyclonal serum to the same antigen in both guinea pigs and rabbits with the aim of using them as capturing (guinea pig) and detecting (rabbit) antibodies in inhibition ELISA format. However, the rabbit and guinea pig sera cross-react with each other. I have even tried using negative rabbit and guinea pig serum, but they too cross-react. Is this a common observation? I can't seem to find any papers where the two species have been used together in inhibition ELISA format successfully.
