I am interested to know the Cr species in NaOH post desorption. The DPC method works only in an acidic condition. The addition of HCl: water (1:1 ratio) of pH1 is thus included in the protocol. Will the presence of NaOH affect my result? OR is it negligible as the addition of acid neutralize NaOH and possibly acidify my solution as desired? Can I still go ahead with this protocol (DPC)?

Your question has the answer hidden in it.
Note that, if DPC method works in acidic conditions as you mentioned, therefore when you add HCl:Water (1:1) solution it will neutralize the pH to nearly 4-5 (you should check the pH after addition). Now you want more acidic condition to perform the DPC method so, you should add a little more aqueous acid to lower the pH to near 3.
Be careful in pH adjustments.

Surely it would be safer to chromatographically separate the species in the alkali then measure them separately ?
Any CrVI present in the NaOH will be compensated for in your reagent blank. You have to run a reagent blank through the entire analytical process; this will correct for the presence of trace amounts of CrVI in your reagents.
