I have formed Giant Unilamellar Vesicles (GUV) via electro-formation. The DLS results do not always match what I see under a microscope (usually between 3 -10 microns) and that made wonder if DLS size distribution results are reliable enough for Giant liposomes.

At particle sizes greater than 1 micron, DLS measurements become increasingly sensitive to a number measurement artefacts which may be mitigated by careful sample preparation.
Are your DLS results repeatable and did you instrument give any indication of data quality? I would recommend looking at this before trying to compare with another technique, but in general at these sizes laser diffraction would be more suitable.
I don't think DLS is suitable for measuring size of GUV of 3-10 micrometers. SEM is a better option
I agree with the previous comments. Also take into account the size distribution you select for the DLS analysis; microscope is usualy expressed as number distribution (frequency), though sometimes it is not said.

If you have a monodispersed solution for GUV with suitable PDI, you can use DLS and refer to it somewhat (be careful about PDI after measurement); however, if you have accessibility for SEM and TEM (is better than others), I recommend to use these microscopy methods.
