We have ACQUITY UPLC Peptide BEH C18, 130Ao,1.7um, 2.1*150mm
for Peptide mapping.
The mobile phase which were tried
1) A: 0.045% H2O, B:.045%ACN
2)A: 0.045% H2O, B:.045%ACN
Various gradient were tried using this mobile phase.
The UPLC was attached to QTOF system
I use commercial CytoC digest as a standard for testing my HPLC systems, 1pmol for nanoLC and 8 pmol for capillary LC. My mobile phase is usually 0.1% formic acid for both A and B solvents, and typical gradient is 3-15-50%B followed by column wash and equilibration. I do not see why this standard would not work for UPLC, maybe a linear gradient 3-50%B will work better due to super short times on UPLC. Hope it helps.
Neelesh, given that you have a Waters Acquity UPLC, I'm suspecting you are using the Waters Cytochrome C digest standard? This is a suitable standard for testing UPLC performance. The care-and-use guide from waters (https://www.waters.com/webassets/cms/support/docs/720004341en.pdf) provides an example of separation conditions on an Acquity UPLC using a shorter version of the column you have (Page 3). This perhaps is a good place for you to start. Otherwise, I am confused by your mobile phase description. I would doubt you are using 0.045% water for A or 0.045% ACN for B, so perhaps there is a typo in your description.
