I am interested in protocol for doing poliovirus neutralizing antibody assay in human samples of serum, plasma or PBMC culture. If anybody has a protocol can you please share it? Where did you buy poliovirus? is it from WHO? do you have to grow your own virus stock?

Kindly see the document attached herewith for polio neutralizing antibody Test. I hope it will help you as most of the Polio Reference Laboratories are following the same with a few modifications.
From late 1950s to early 1970s, Dr. Albert Sabin distributed his strain to manufacturers, in 1972 he donated his strains to WHO to manage the distribution of viral seeds for vaccine production.Master seeds were prepared from original virus produced by Sabin and were described as SO+1 and further working seed lots were prepared and designated as SO+2 .WHO is the custodian of Sabin master seed for type 1,2 & 3.
Sabin reference strains are available at NIBSC, UK for procurement.
Regarding the virus stock, working seed lots are prepared from either master seed or reference strain following seed lot system taking care not exceed the maximum allowable passage number, hundreds of aliquots are prepared and stored -20 freezer for future use.
the 1997 version has been replaced bij the 2004 version. be aware that in 2016 OPV-2 vaccination will cease and handeling of OPV-2 should be under BLS 3conditions
