I've heard that a CMV promoter is not usable in myeloid cells, because of methylation and subsequent silencing. However, I can't find some background on this.
Can anybody tell me if it is ok to use a CMV promoter in a lentiviral vector for myeloid cells?
I have used a pCMV promotor in lentiviral constructs transduced into myeloid cells with no problems. No idea about a more general overview though im afraid!
You are the first one to tell me about a positive result... I only have heard so far that a CMV promotor gets silenced in myeloid cells..
Ive used lentiviral constructs with CMV in the myeloid cell lines Kasumi-1, HL-60, NB4, MV4-11 and U937 with no problems and have done long-term culture of HL-60 and Kasumi-1 with effective expression (with ectopic gene expression) or knockdown (via inducible shRNA). Im pretty sure there are some published data, especially with the U937 line too.
