I am looking for an antibody for HSP beta1 (small heat shock protein in Crassostrea gigas EKC40046.1). In mouse HSP beta1 is also known as hsp27, hsp25 (UNIPROT kb/SWISS-PROT: P14602.3).
I aligned HSP27 of Crassostrea gigas with that of mouse and I found 44% query cover and 40% identity. Is this percentage good enough to select an antibody from mouse for identifying HSP 27 in C. gigas?
I found in literature that many researchers have detected HSP 70 in Crassostrea gigas using an anti HSP70 antibody from mouse (where the identity between the 2 was 73%).
You need to look in detail on the regions of the epitope ( say 10-20 aa) where there is perfect match. Check whether the cross species antibody recognizes those 10-20 aa which are similar between the two species at protein level. In such a case the overall sequence similarity may not matter but only the regions of similarity that are recognized by the antibody raised in different species.
It all depends on whether the right epitope is covered in the 44% sequence that you have got otherwise it is not sufficient to go for the Ab
