I am looking for methods and targets on which one should look at if we are studying a molecule's role in maintenance of synapse for long- term after certain activity is induced. Let's say if it increases synaptogenesis, but how to study if that molecule facilitates maintenance of those synapses for long term.

I was thinking if one can show that synapse are increasing through SNAP- 25/ synaptophysin and PSD-95 immunofluorescence and then if it can be checked in synaptosomal extracts on western blots (IP) for interaction of different adhesion molecules after long time points with the molecule in study and in its absence, that would provide me clues that the molecule in question is able to interact with such synapse stabilisers and hence it is able to maintain synapses for long term.

Please suggest if I am thinking in the right direction or there are other important aspects to look at?
