Currently, I am trying to perform the Intron 22 Inversion for the FVIII gene. I have tried both long-distance (LD) PCR and Inverse Shift (IS) PCR, but I was unable to get the results in either way. In the case of long-distance PCR, I am using highly purified fresh samples, but I am still unable to get any bands. Can anyone help me in rectifying or standardizing this reaction? Is there any other protocol to establish this?
