I do not know exactly if I need to make a STD curve every time I make an enzyme activity test.
Also, I am not sure if I have to take in account the substrate dilution on the final enzyme activity equation.
I would be greatful if you could help me and recommend me some related articles.
If you are following a standard operating procedure (SOP), and it includes preparation of a standard curve, then you should do it.
For more informal situations, it would still be advisable to make a new standard curve each time because your detection reagents may not always be identical. However, if your experience is that the standard curve never varies significantly, it would probably be OK to use a previous standard curve.
If the reagents you are going to use in each assay are being taken from the same lot, you can use the same glucose standard.
For every new assay, you must prepare a fresh standard as there might be a number of varying factors that can affect the assay.
Personally, I prepare standard curve for each assay. I learnt the hard way not doing so. This is one of virtues of a good researcher - Perseverance.
