I do not know exactly if I need to make a STD curve every time I make an enzyme activity test.
Also, I am not sure if I have to take in account the substrate dilution on the final enzyme activity equation.

Preparing the standard curve depends on the time interval of your test and the number of using the spectrophotometer. If time interval is more than one month or the spectrophotometer is constantly used by different people , it is better to prepare standard curve every month.
Whenever the substrate is diluted, it is necessary to concider the concentration of substrate.
It is usual to recieve different respond and reaction against different substrates for same enzyme. But, the enzyme activity depends on type of substrate. So, it is necessary to mention its activity base on used substrate.
