I am working on Liposome and want to Exchange/ replace the external buffers with oily vesicles. So to initiate that process which are the basic methods or techniques to carry out this process and what are the major process parameters which can be affect the process.
Mihir can you clarify whether you intend to pack desired material inside the liposomes or adsorb outside? The liposomes may be made up of phoshatidyl choline and cholesterol. What's the purpose of making liposomes?

Its a novel drug which will be working as an anti-cancerous agent and the material or the oily vesicle which will be used to exchange the external buffer only for adsorb outside.
Basically the API is in a specific buffer and I want to exchange that buffer with my desired oily vesicle solution where my molecule is quite stable.

If you intend to pack the drug inside, the method is bit different for making such liposomes!

Just to add some points on it, the liposome consists of that anti cancerous agent which is stable in that oily vesicles. so I want to exchange that buffer to oil emulsion and keep it stabilized. So which techniques are the best suit for Diafiltration? Ultrafiltration i.e UF/DF or Gel filtration chromatography?
Actually the statement ' I want to exchange that buffer to oil emulsion and keep it stabilized' you are making is confusing. If you have prepared liposomes with drug entrapped in them, simply centrifuge these liposome, they must sediment because of their weight/ mass. The settled/ sedimented liposomes may be kept suspended in 0.05 M Phosphate buffer pH 7.2 containing 0.002% (w/v) thimerosal as a preservative.
