I used InVivoMAb anti-mouse Ly6G (Clone: 1A8) (bioxcell) to deplete the MDSCs in balb/c mouse. However, after one dose of 200ug, all the mouse became obviously sick, and one mouse died 3 days after the injection. I used the antibody according certain published articles. Then why would this happened?
Did you include isotype control as well? how are those mice looking? Also more experimental details will be helpful to trouble shoot the issue. how old are the mice? route of administration? buffer ph?
Sunagar Raju Yes, I did use IgG isotype control. And this group of mice looked fine. Mice were 5-6 weeks old. 200 ug anti-body diluted in 100ul PBS (PH 7.2) was injected i.p.

For my understanding you are depleting neutrophils and partially monocytes (a bit of cross reacitivity on Ly6G/Ly6C). Those cells are the first line of innate immunity defense against infection. Your mice may I got sick and died from it.
My second hypothesis was that your antibody was with preservatives (such as sodium azide) which can kill the mice, but from the purchaser website it should not be the case
