In vitro micronucleus assay using CHO cell procedures?

I'm doing in vitro micronucleus assay using CHO cells, ethanol : acetic acid (3:1) as fixation solution and Giemsa 5%. I also did centrifugation at low speed and pipetting gently but why did I still have fragmented nucleus? And the color of the cells is not intensive. Anyone can help?
