Does anyone have experience with the in vitro generation of monocytic MDSCs or MDSC-like cells from isolated human monocytes? If yes, could you share your experience on this?
After some searching I came across a study by Heine et al. (2017) PMID: 28507805, in which monocytes are incubated with IL-4, GM-CSF and IL-10 for 7 days. The following analysis of the resulting cells, which the authors call IL-10-APCs, revealed cells with phenotypic (CD14+, HLA-DR low) and functional characteristics comparable to monocytic MDSCs (immunosuppressive when co-cultured with moDCs or T cells).
What is your opinion on this protocol? Do you have suggestions/remarks, whether this protocol makes sense? I have also come across another protocol in which monocytes are incubated with GM-CSF only for 3 days to generate so-called licensed monocytes and are subsequently stimulated with IFNg and LPS to generate MDSC-like cells. Does this make sense in any way?
I would appreciate any comment or suggestion as I am very interested in generating MDSCs in vitro. Thanks in advance!
