Samples were swirled well in the solvent followed by centrifugation at 10,000 g for 10 min. The supernatant was stored on ice for SA measurement. 100 μl of the supernatant was mixed with 0.1% freshly prepared ferric chloride. The volume of the reaction mixture was made up to 3.0 ml and the complex formed between Fe3+ ion and SA, which is violet in colour was determined by spectrophotometry, measuring the absorbance of the complex in the visible region (at 540 nm). :
I want to know:
1- quantity of 0.1% freshly prepared ferric chloride which should be added to the supernatent 2- The type of solution is used to made up the reaction mixture volume 3.0 ml
