output request should be for reference point set or point of application set?
how to give the equation constraint , what's the significance of putting -1 , 1 etc

I am not fully sure about your questions, but you can try to have the reference as a point set and an application set and see the difference in results. Also, what you mean by the equation?

the constraint equation you set up should always equate to zero. A simple example would be to set the x-displacement of Node 1 equal to that of Node 2:
u_xNode1 - u_xNode2 = 0
Therefore, the first node would have the sign factor 1 and the second -1.
As for the output request you can define a set in Abaqus and request special History and Field Outputs for that set.

Hello. The previous answers addressed you last 2 questions. Concerning your first question, you can apply a displacement to a beam via the boundary conditions tool. Rather than constrain motion, you can specify a reference displacement directly in the boundary conditions window (or in the boundary conditions definition section in the input file). ABAQUS will apply your displacement-based load as it would with a load, depending on your analysis of choice (static, Riks, etc.).
You can apply the displacement in the place of loading. The reaction force of supports will be your output for force and if you define a Set for loading point, you can get the displacement.
