I have to find IC50 value for my metal complexes in MCF-7 cell line in 24 hours time point. If I want to compare with previous papers, most of the papers find out the IC 50 with 72 hours and even 96 hours. When see that metal complexes concentrations which are less than 1 micromolar and nano molar.
To show the very less concentration have they done the experiment in that time point? If no, universally, what is the correct time point to find the IC 50 value?
you can do a IC50 in 24, 72 and 96 hours, End the final of all hours could do IC50 among all times, this way you have a mean of IC50 around the time.
There is no "universal" correct time point to find the IC50 value of a given compound.
If you want to determine the growth inhibitory concentration (GI50), you have to do it at about one time the doubling time of the cell line of interest, and about two times the doubling time.
In cell culture, the large majority of adherent cell lines display doubling time between 18 and 24 hrs. The reference "lab" for analyzing GI50 concentrations is the NCI (USA - Bethesda - MD). The NCI use 48 hrs of incubation with the drug of interest on their 60-cell-line panel. In my research unit, we prefer to use 72hrs.
24 hrs is too short to determine a reliable GI50 concentration.
In contrast, if you want to characterize the mechanism of action of a given compound, let say "compound X" that kills 50% of the cells of a cell line of interest (say "Y") either at 48 or 72h, you have to use this Y concentration of compound X to determine its IC50 concentration at several time points before 24hrs to prove that the time point you choose to characterize the mechanism of action of X is not too toxic.
