I am testing a new antibody (CD94) to stain for NK cells in the brain. However, when I test it in tonsil tissue, it stains basically every cell in the germinal center, so I did another test with CD20 for B cells, and the CD20 is also in every single cell in the germinal center (even when using confocal and creating a high resolution Z-stack).
At first, I thought maybe the antibody is aspecific. However, when I use it in white matter lesions of MS, the CD94 and CD20 both stain some cells and show zero overlap. Therefore, I guess it has nothing to do with the antibody but more with the tissue.
Does anyone have experience with staining immune cells in tonsil, and is it normal to see overlapping immune cell markers that shouldn't be on the same cell?
