Bacterial lipases are greatly influenced by nutritional and other factors, such as temperature, pH, nitrogen and carbon sources, inorganic salts, agitation and dissolved oxygen concentration. So, these factors to be taken care while testing any bacteria for lipase activity.
The ability of organisms to hydrolyze fat is accomplished with the enzyme lipase so, this test was conducted to study the ability of PHAs producing bacterial isolates to hydrolyze lipid by the action of enzyme lipase. Thus, tween NA plates were prepared reducing the concentration of nutrient broth to five times. Then 1% [V/V] tween-20 and 1.5% agar was added to the culture medium as lipid and solidifying agent respectively. The isolates were spot inoculated on the plates and incubated at 37oC for 24 hours. Moreover, plates were then stored at 4oC for 2-3 hours to precipitate the lipid in presence of CaCl2. A clear zone around indicated positive lipolytic activity of the isolate caused by the production of lipase.
