Hii, I am working on telomerase enzyme and accidentally kept the protein lysate at room temperature for about 24 hours. Telomerase enzyme is heat sensitive but since my samples were kept for 24 hours at room temperature so will it affect the activity of telomerase enzymen?
Proteins vary widely in their tendencies to lose activity during storage. It would not be surprising if an enzyme sample lost activity due to storage for a day at room temperature. You used the phrase "protein lysate," which suggests that your sample was a crude extract. Because crude extracts usually contain proteases that can cause protein degradation, and can also be a good medium for the growth of bacteria that can secrete proteases, I think it is very likely that your sample will have lost all or most of its activity.
Even if you do get some activity of your enzyme, the question you have ask yourself is would you trust the data generated from this sample to be of value (assuming that establishing different ways in which an enzyme can be stored/abused is not the main goal of your study)? ;)
