As already mentioned by Max Anstötz doing morphometry for neurons and specific to granule cells of DG not be easy to mix. However, in my opinion the best and relatively easy option for you could be doing Golgi staining alongside Prox1 immune histochemistry. You will find many available protocol for immunestaing along with Golgi im[pregnation. In this way you can target the granule cells (through Prox1) and can also do morphometry of the neurites (tracing banches form Golgi impregnation). The other way to replace Golgi-impregnation as it is sometimes hard to do the both (as it is metal impregnation) you can obviously replace Golgi with Dil.

The other option is to directly go for lentiviral transfection. However, it will again not be that easy in in-vivo condition. For slice culture is could be easy and effective.

Hi Nada,

You can try Golgi-cox staining for dendrites, soma, and spines.

But the axon is a bit tricky because it's tiny and crosses different levels (especially on brain slices).
