gene a have many homologous genes. all found in the same position. note: one of homologous have the same function of this gene but gene a didn't work at high temperature. and our homologous gene do the same function at high temperature.
if we want know the sequence of the homologous gene which not cloned before ... only what we know the sequnce of the origin gene (a) and that our gene one of its 6 homologous genes. but the only of the six that do the same function

In principle, one could clone a long-range high-fidelity PCR amplicon of all your homologues into a vector, transform it into a microorganism and sequence all of the clones.
Thank you Dr. Roxana for attention . i will work on plant. you mean that by knowing of original gene which is already sequenced. i can design primer for PCR then cut the gel bands for sequencing then i can check its function
i will illustrate what i want . the gene is (A) its function is resistance in certain temperature if the temperature is high , this gene will not work. but in another line of this plant , they found that there are resistance at high temperature. this is because found of another gene called (A1) they found that this gene found in the same chromosome of gene (A) which is already sequenced. also they found that there are another homologous genes (A2 A3 A4 A5 A6) to the gene A but the gene A1 is the only homologous that have resistance function but at high tem. in one line of this plant. so i mean that can i isolate DNA from this line which contain A1 and others gene A2 A3 A4 A5 A6 then done PCR by A primer then from the gel , i will sequence product then check its function to know which one of them is A1 or not?
