If you are talking about Saccharomyces cerevisiae then most likely not since it is known to have very little introns. You are better off using cDNA.

Hi I agree with Hanna (and depends on which yeast), it is normally advisable using cDNA. Another good reason is is also easier to transform, since is shorter than the original gene (2 introns less)
