You'll prob need to concentrate your bacterial inoculum and harvest spores for Aspergillus. Clean up incoulum - spent broth will be confounding. Dilution plating will tell you ~ viable count going in.
I concur with Phil's comments. That is heavy innoculum, are you really sure you want that much? With bacteria that will be noticeably turbid culture. I'm not sure you will be able to get that high with Aspergillus, but someone who works on it can correct that.
But you will need to just titer the innoculum after you make it in order to confirm that you have the numbers you need.
