I am planning to stain floating sections of mouse brains that were fixed in 4% PFA and then kept in PBS.
The mice had undergone hypoxic ischemic brain injury as neonates, and sacrificed as adults (30 days later). I will be doing Cresyl violet (Nissl) and Iron (Perl's) stain, but apart from that, I would like to also look at the demyelination. Are any good antibodies/ staining methods I could use? Would TUNEL be useful 30 days after the damage?

Hallo Alkist, I think the Nissl-stain will give you alot of informations. Helpful will be alsinformations about the gliosis (Immunhistochemistry with GFAP; DAKO) and the demyelination (IHC with MBP = Myelin Basic Protein; Millipore MAB382). I am not sure weather TUNEL would be helpfull becaus at that time most of the demaged tissue will be replaced by glia fibers.
