Hi All, Recently my colleague did an MTT assay on the effect of Doxorubicin on Hela cell line and I found that The inhibition after 48 hours is less than of after 24 Hours!!!. what happened? Anyone could help me to find what is the problem or if that is correct?

important points to be considered, such as ( shelf life of the drug, and cell proliferation rate).
if you cells are proliferating every 24 hr, then your drug concentration is getting diluted by cell division after 48 hr, which explains that 48 hr has lower effect as compared to 24 hr incubation.
