You can use this protocol for conversion of TAG into FAME.
For methanolysis of the fatty acids, approximately 10 mg of freeze-dried cells were weighed and placed in Teflon-stoppered glass vials, then 1 mL
chloroform and 1 mL methanol/H2SO4 (85:15 v/v) were added to
the vials, after which they were incubated at 105 C for 2 h, cooled
to room temperature, and then incubated on ice for 10 min. After
adding 0.5 mL of ice cold water the samples were thoroughly
mixed by vortexing for 1 min, and then centrifuged at 2000g. The
organic phases (bottom) were extracted by pipette and moved toclean borosilicate glass tubes, containing Na2SO4. Use the sample further to perform GC MS analysis.
You can use this method for all type of oils containing fatty acids.
