Does anyone who used poly RGD know where to find any? Should I ask Sigma or similar distributor to make a custom long polypeptide? Will coating the plate with RGD instead of poly RGD work in term of neutrophil Activation?
There are a number of ways you can activate integrins. You do not necessary need poly RGD. In theory RGD by it self will activate integrins even though a poly RGD will both activate and cluster them... And I think in the field there still is some discussion on what is the most important step to trigger signalling. Anyway, I believe the easiest (and may be cheapest) way to activate the RGD binding integrins is probably by coating your plate with Fibronectin. Of course coating with RGD will work as well. You can also somehow modulate your stimulus by changing the concentration of your coating reagent.
thanks, I usually use fibrinogen to coat the plate but I wanted a more potent stimulus to activate neutrophils without any other stimuli. I understand from your answer (RGD activates but poly rgd also clusters) that I will need poly RGD. Do you know if it's commercially available?
