I want to know you are using CASA to analyse or evaluate semen, if you use which one you use (CEROS, Androvision, ISAS or others). And what kind of problems have you faced when using CASA? Can you say your problems are about hardware or software? Also do you trust the data from CASA?
You will have to standardize its basic parameters/standard values in CASA software to (which is species specific too) get the results for sperm parameters. Proper dilution and loading of its slides is necessary too. Yes definitely you can trust its values if your hardware and software both are working well.

Thank you for your answer, It is well know that standardization and calibration is an important factor affecting results of CASA. Although you make all standardization and calibration for semen of currently analysed species, can you trust the results when compared with your subjectif analyes(i.e. subjective motility)? Moreover, proper dilution and loading of sample to chamber are needed extra-time and cost. Can you use standart slipe and cover-slipe for evalution of sample in CASA?
I have used 12 well multi-test slide (ICN Biomedicals Inc.) with standard coverlips when using CASA. Although I did not work on mammalian sperm so I am not sure if it would suits your needs. Regarding the validity of the data, you will need to do extensive stats to check the data distribution, etc to see if the data is solid or not. hope this helps.

yes I did, the distribution of the data looked really nice. The multiwell slide provides a "mini-chamber" effect and you can coat your coverlips with 1% BSA (Bovine serum albumin). Please note this method has been used for fish and amphibian sperm, but not mammalian.

I'm technical manager of microptic (developer of the CASA system Sperm Class Analyzer - www.micropticsl.com) and if you want to have accurate count, is better to use disposable counting chambers. Of course you can use other kind of slides but the results will not be accurate. As Severine said, if your system is calibrated, use a quality microscope and all the configuration well set, the results will be accurate and precise. And not only for concentration, but also for motility, morphology, dna fragmentation and vitality.

Thank you for your information about CASA. It is the best way to use disposable chambers to gain true results. However, it causes additional expense for each analyses. I saw a chamber developed by Proiser. It can be used alot of time (clean and use). Nevertheless, the price of chamber is approx. 250 € (I think it is expensive compared with its function). I think conventional slides and covers is best way for decreasing expenses. Or CASA developers should develop cheap chamber systems...
