I want to extract different compounds (organic acids, 3-phenyllactate and other small peptides ) from bacterial culture. I have made some extraction fractions by solvent (methanol, ethyl acetate and dichloromethane) extraction now I want to extract/purify that particular fractions by silica gel chromatography. What would one can recommend me the pore size of silica gel for compounds written above?
like: pore size ~0.8 cm3/g pore volume, 60 Å
That sounds like the standard flash chromatography media, especially the 60 Å pore size. That will work for those compounds. Column packing, pH, and so forth will probably play a bigger role in your purification than the pore size and volume. The peptide scientists discuss the role of pore size in purifying those, relatively large, compounds.
@Jack Silver: If I talk about peptides I forget to tell one thing that is cyclic dipeptides (small peptides), In this case what should be the pore size of silica gel.
Don´t use silica gel for first purification, try Biogel P2 with water which works well without adsorption problems.
@Muhammad- I still would recommend using the standard silica for cyclic peptides. Also consider alumina for cyclic peptides, with elution with methanol. Note that many natural products are a similar size as peptides, yet standard silica works well for them. The peptide chemists are divided about whether using wide-pore C18 is advantageous.
