Ethyl acetate extract has no antimicrobial activity. It is also based on the polarity one of the solvent system normally used. The minimum antimicrobial activity of the solvent has to be ignored. More over a few compounds only dissociated with ethyl acetate and you do not worry about this particular issue. Thanks and regards Mr.Muhammad Numan.
Yes..All solvents should be removed from extract before further work. The stability of extracts with remnants of solvents may be compromised. Dry weight is also required in standardisation of any bioactivity. All the same, .incorporate whatever extraction or reconstitution solvent used as the negative control. The negative control should ideally not exhibit any appreciable bioactivity.

I did not test the antimicrobial activity of ethyl acetate but I think that this solvent can disrupt cell membrane (to be more precise the phospholipid bilayer) in direct contact with the cell. The alcohol acts in the same way for example. I think this solvent has antimicrobial activity.
Ethyl acetate is the solvent that we use in the extract. Yeah, as such ethyl acetate can have minimal antimicrobial activity since it disintegrates the phospholipids present in the cell membrane of bacteria. So before you assess the antimicrobial effect of any extract, solvents should be removed. The best way would be using a rotary evaporator. After removing the solvent, you can resuspend it in a buffer solution. However, a fractional action of solvent will still be present. But this can be standardized by using these solvents as your negative control.
Use well diffusion method to check the antimicrobial activity. Use on well as negative control which will contain pure ethyl acetate solvent. Remember use glass Petri dish only. Dont use plastic petridish for this purpose otherwise plastic may melt by ethyl acetate. Thus, you can see the difference between zone of inhibitions of pure ethyl acetate and your extract.
