if I differentiate THP-1 cells using PMA and later in the experiment if I change the medium of THP-1 cells to xombination of MEM and RPMI, what is the possiblity that cells will dedifferentiate?

Dedifferentiation is not a common occurrence in THP-1 cells after PMA-induced differentiation as this process is more complex and influenced by various factors. While PMA withdrawal can lead to dedifferentiation, it is not an automatic or guaranteed outcome.
Instead, you should focus on ensuring that the combination of RPMI/MEM medium supports both cell types and monitor the culture for any unforeseen issues. I feel a mixed medium of RPMI and MEM can still support both HepG2 and differentiated THP-1 cells, as long as it provides the necessary nutrients and growth factors.
