Are you using cell lines or tumor samples from mice or humans? Because in my hands, only fibroblasts from patients are attached, and tumor cells grow as oncospheres.

If this is not your problem, may be is the FBS in your culture media. I never use FBS in an oncosphere formation assay.

Another question, do you use commercial low attachment well plates? The ones from corning are the better ones.

i use cell lines and i used before low attachment plates with tumorsphere special medium but the cells keep attaching to the plate.

recently I used ultra low attachment plates from corning and it gave me great results.
