Sample Scoring
A-1 1 0 1 0 1 - 1 - 1 --> polymorphic band
A-2 0 0 0 0 0 - 0 - 1 --> unique band
A-3 1 1 1 1 1 - 1 - 1 --> monomorphic band
A-4 1 0 0 0 1 - 0 - 0 --> polymorphic band
I have 9 samlpes with 1primer. There is no amplification for 2 samples hence depicted as - . Please, tell me , have i taken the polymorphic, monomorphic and unique bands correctly?
Hi, I don't understand this scheme. I know that 0 means no band and 1 means there is a band!
Hi Martin & Nadine , Yes i have 9 different samples and primer A has shown 4 bands (loci) i.e. A-1, A-2, A-3, A-4 . Sample no. 6 and 8 are not showing any of the 4 bands. i was told that as there is complete gap, the sample DNA is not amplified with the particular primer. In light of this my 2 questions:
1. if i should give 0 score to these gaps ?
2. if i give 0 score to these gaps ,then A-3 band will be polymorphic? In fact if i dont consider the (not) amplified sample score , then the band seems to be monomorphic.
for Nadine, i repeat the score again.

Hi, did you test sample 6 and 8 to make sure that the DNA/cDNA was intact (and then everything worked proberly)?
Hi, I got it amplified very nicely with other primers. that means DNA is of good quality. so i tested it twice after 10-15 days for that A primer, but got the same result i.e. complete gap at 6 & 8.
