I am doing hydrolysis and solubilisation of starch. The method I am following states that Pancreatic a-amylase measuring 4.0mL containing 3 U/mL of amyloglucosidase solution was added to each tube.
In my possession I have a >260U/mL amyloglucosidase. What should I do?
Take 1.15ml of the 260u/ml and dilute to 100ml volumetric flask. Or take (3V/260)ml of the stock solution and dilute to V (where V=volume of your flask)
As Adugna has noted, volumetric dilution will be the answer if you have quantified your current concentration.
What quantity in ML of 3U/ML do you need?
Once you know that volume use the formula V1C1=V2C2 where V1 is the volume of 260 U/ML you will need to take from 260 U/ML, V2 is the volume of 3 U/ML needed for your experiment, C1= 260 U/ML and C2 = 3 U/ML. On ice, pipette V1 into a sterile tube and top up to V2 with sterile water. V2 will be your requires quantity of 3 U/ML of AMG.
