I am wondering why to monitor mesenchymal stem cells proliferation we pick up days 1, 3 and 7. I believe that it has something to do with the fact that the log stage of cell growth happens between 1-8 days. However, I have heard some people talking about doubling time as well. I am a bit conduced about how to find a connection between the 3 famous days, log stage and doubling time.

For U937 cell line and HSC, the doubling time is near 22 hr, but it started after a lag time after the initial seeding. Therefore 3, 5 and 7 is sufficient time period.
Thank you Younes, so if it is 22 hours, how you figure out the picogreen days(3, 5 and 7). My professor believes that the first assay in mesenchymal stem cell should be done 6 hours after initial seeding to check how even cells are distributed and if they are alive and attached to the surface. Then the general trend in our lab is days 3 and 7. But I am not sure how we come up we this days. How you used doubling time to decide your days?
