Please start with tender parts of either internode or leaf with midrib but cut lamina as explant(s) and BAP and NAA as preferred cytokinin and auxin. It does not matter whether the plant is indigenous to Nepal or else where, what matters in PTC is that whether the plant is a tree, shrub or herb, whether it exudates phenolic substance or it is difficult to decontaminate.Please consider all these factors before starting TC of a new plant species. thanks for the comment, these are all set up but i am not getting what are the factors that i need to consider for getting the callus, usually hormones are done but why only the MS media , are there any supplements to be added. I am to perform the tissue culture of new plant that is indigenous to Nepal. Thanks

Try to find taxonomically related species where tissue culture has been made. Although the composition of media is strongly genotype-dependent (even within species), this may give you good suggestions on what could be the best media for you.
1. Make a literature survey on the plants related to your species. This will give you some guidelines.
2. What is your aim? Plant regeneration or just a callus culture? What type of tissue culture do you intend to use- callus culture, micropropagation? This will influence medium composition, hormone content of your media.
3.What type of explant will you use? This is important for choosing a proper surface sterilization method.
