Hello,

I am trying to perform a FISH with painting probes on a cell population selected by FACS but there are problems with the hybridization.

The cells are fixed and detected by an indirect immunofluorescence. The interest populations are then isolated using a cell sorter. The problem appears when I perfom the FISH and there is no hybridization. I have tried various fixatives and protocols but non of them seems to work. The probes work well because the control hybridized.

I have searched for a sequential cell sorting-FISH protocol but I could not find much information.

Does anyone have any suggestion?
