I have to do a competition growth curve with two different isolates (Salmonella and Lactobacillus). What approach would you use to do this?
I thought I could grow them seperately (Salmonella in LB and Lactobacillus in MRS broth), add them both into the media I choose and take optical density readings, also, to determine if one has inhibited the other, I could then plate them onto LB agar and MRS agar and record CFU.
Does this sound right?

To prepare a competitive growth curve you need to culture both organisms simultaneously and need to monitor growth of each on a time scale. Best shall be to count cfu after appropriate dilution of mixed culture using a selective medium instead of OD at 600 nm. The latter will reflect combine growth but not the growth of individual bacterial strain. You may monitor lactic acid concentration simultaneously as well as Salmonella somatic antigen, if possible!
